• Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at room temperature for 20 min.

Target Information

DNA-binding protein SATB2 binds to DNA at nuclear matrix- or scaffold-associated regions.
It recognizes the sugar-phosphate structure of double-stranded DNA and acts as a transcription factor controlling nuclear gene expression by binding to matrix attachment regions (MARs) and inducing local chromatin-loop remodeling.
SATB2 also serves as a docking site for chromatin remodeling enzymes, recruiting corepressors (HDACs) or coactivators (HATs) to promoters and enhancers.
It plays a key role in the development of upper-layer neurons and regulation of corticocortical connections in the cerebral cortex.
Mutations in SATB2 are associated with isolated cleft palate (CPI).
