Product Specific Information
- For PFA-fixed tissues: incubate antibody for 30 minutes at room temperature.
- For formalin-fixed tissues: heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes.
Target Information
SATB2 is a DNA-binding protein that associates with nuclear matrix or scaffold regions and recognizes the sugar-phosphate backbone of double-stranded DNA. It functions as a transcription factor regulating gene expression by binding to matrix attachment regions (MARs) and inducing chromatin-loop remodeling. SATB2 acts as a docking site for chromatin remodeling enzymes, recruiting corepressors (HDACs) or coactivators (HATs) to promoters and enhancers. It plays a crucial role in cortical neuron differentiation by modulating BCL11B expression, repressing Ctip2, and regulating corticocortical connections. Mutations in SATB2 are associated with cleft palate isolated (CPI).
