• Immunohistochemistry (PFA-fixed): Incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

DNA-binding protein SATB2 binds to DNA at nuclear matrix- or scaffold-associated regions. It recognizes the sugar-phosphate backbone of double-stranded DNA and functions as a transcription factor controlling nuclear gene expression by binding to matrix attachment regions (MARs) of DNA and inducing local chromatin-loop remodeling.
SATB2 acts as a docking site for chromatin remodeling enzymes by recruiting corepressors (HDACs) or coactivators (HATs) directly to promoters and enhancers. It is essential for initiating upper-layer neuron (UL1) genetic programs and repressing deep-layer (DL) and UL2-specific genes, likely through modulation of BCL11B expression. SATB2 also represses Ctip2 and plays a regulatory role in corticocortical connection development. Mutations in SATB2 are associated with isolated cleft palate (CPI).
