Target Information

F-Box and WD Repeat Domain Containing 7 (FBXW7) is a member of the F-box protein family characterized by an approximately 40 amino acid F-box motif. These proteins form part of the SCF (SKP1-cullin-F-box) ubiquitin ligase complex, which mediates phosphorylation-dependent ubiquitination.
F-box proteins are categorized into three classes:

  • Fbws: containing WD-40 domains
  • Fbls: containing leucine-rich repeats
  • Fbxs: containing various interaction modules or no recognizable motifs

FBXW7 (also known as FBX30) belongs to the Fbws class and includes seven tandem WD40 repeats. It binds directly to cyclin E and is thought to target cyclin E for ubiquitin-mediated degradation. Mutations in FBXW7 have been identified in ovarian and breast cancer cell lines, suggesting a potential role in tumorigenesis. Multiple transcript variants encoding different isoforms have been identified.
