Clone M053 mouse monoclonal antibody detects a 110 kDa protein on SDS-PAGE native or denatured immunoblots of human THP1 monocytes. The antibody prefers the native CD244 protein but also detects denatured CD244. Suitable for western blot, immunoprecipitation, immunocytochemistry, and ELISA capture.

Target Information

CD244 (Natural killer (NK) cell receptor 2B4/SLAMF4) is a signaling lymphocyte activation molecule (SLAM) receptor belonging to the Ig superfamily. It has two Ig-like extracellular domains and a cytoplasmic domain containing four immunoreceptor tyrosine-based switch motifs. CD244 binds to CD48, a receptor expressed on hematopoietic cells. Its activity is regulated by adapter proteins SH2D1A/SAP and SH2D1B/EAT-2. Downstream signaling involves VAV1, INPP5D/SHIP1, and CBL. Activation of CD244 enhances NK cell cytotoxicity, IFN-gamma production, and granule exocytosis. It regulates CD8+ T-cell proliferation and modulates inflammatory responses in dendritic cells. In cancer, CD244 expression increases in intratumoral DCs and myeloid suppressor cells, and anti-CD244 therapies may enhance T-cell infiltration and inhibit tumor growth.
