Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. Natural serum levels of IgE are low, representing a small fraction of total immunoglobulins. IgE mediates allergic responses by binding to mast cells and basophils via the Fc epsilon receptor I, triggering histamine release and other mediators. IgE21 recognizes both soluble and membrane-bound forms of IgE.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells at 5 µL (0.2 µg) per test (100 µL final volume). Recommended cell count: 10⁵–10⁸ cells/test.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer Usage:
Updated conjugates ship with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance. Add 5 µL per sample before mixing with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

IgE is one of five mammalian immunoglobulin classes. It consists of two epsilon heavy chains (each with four constant domains) and two light chains. IgE is critical for type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylactic shock) via Fc receptor binding on mast cells and basophils. Crosslinking triggers degranulation and release of allergic mediators. IgE also contributes to immunity against parasites.
