Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE exists at low levels in serum and plays a key role in allergic responses by binding to mast cells and basophils via Fc epsilon receptor I, triggering histamine release. IgE is produced by terminally differentiated plasma B cells and basophils, and IgE21 detects both soluble and membrane-bound forms.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells at 5 µL (0.2 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample to antibody cocktails before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on engineered nucleic acid dye structures designed to minimize spectral spillover and spread.

Target Information

IgE is one of five mammalian immunoglobulin classes. It consists of two epsilon heavy chains (each with four constant domains) and two light chains. IgE mediates type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylactic shock) by binding to Fc receptors on mast cells and basophils. Crosslinking triggers degranulation and release of mediators that drive allergic responses. IgE also contributes to parasite immunity.
