Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE is present at low levels in serum and plays a central role in allergy responses by binding to mast cells and basophils through the Fc epsilon receptor I, leading to histamine release and other mediators. IgE is produced by terminally differentiated plasma B cells and basophils and exists in both soluble and membrane-bound forms. IgE21 recognizes both forms.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry on normal human peripheral blood cells.
Recommended use: 5 µL (0.2 µg) per test in a 100 µL final volume. Suitable for 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) improves staining performance and should be added at 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL of CellBlox Plus Blocking Buffer per 100 µL of cell sample (10³–10⁸ cells).

Target Information

IgE is one of the five classes of mammalian immunoglobulins, consisting of two heavy (epsilon) and two light chains. It mediates type I hypersensitivity reactions such as hay fever, asthma, hives, and anaphylactic shock, by binding to Fc receptors on basophils and mast cells. Crosslinking of IgE induces degranulation, releasing mediators responsible for allergic responses. IgE also contributes to immunity against parasites.
