Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE is present at low levels in serum and plays a key role in allergic responses by binding to mast cells and basophils via Fc epsilon receptor I, leading to histamine release. IgE is produced by plasma B cells and basophils and exists in both soluble and membrane-bound forms. IgE21 recognizes both forms.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.2 µg) per test in 100 µL final volume. Suitable for 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA-intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample before mixing with cells.
• For single-color controls, use 5 µL of buffer per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates use Phiton™ technology with engineered fluorescent signatures to minimize spillover and spread.

Target Information

IgE is one of the five classes of immunoglobulins in mammals. It consists of two heavy (epsilon) and two light chains. IgE mediates type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylaxis) by binding to Fc receptors on basophils and mast cells. Crosslinking triggers degranulation and allergic responses. IgE also contributes to parasite immunity.
