Product Specific Information
The monoclonal antibody 162 recognizes CRACC (CD2-like receptor for activating cytotoxic cells), also known as CD319 and CS1, a member of the SLAM receptor family.
CRACC is a 66 kDa glycosylated transmembrane protein containing a cytoplasmic ITSM domain (Immunoreceptor Tyrosine-based Switch Motif).
Unlike other SLAM family members, CRACC’s ITSM domain is SAP-independent and instead uses EAT-2 as a signaling molecule. This feature is significant for patients with X-linked lymphoproliferative disease (SAP mutation).
A splice variant lacking the ITSM domain has been reported.
Applications Reported:
The 162 antibody has been reported for use in flow cytometric analysis.
Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended usage: 5 µL (1 µg) per test (final volume 100 µL).
Cell number: 10⁵–10⁸ cells/test (empirically determined).

Target Information
SLAMF7 contains one Ig-like C2-type domain.
Isoform 1 mediates NK cell activation through a SAP-independent ERK-mediated pathway and may play a role in lymphocyte adhesion.
Isoform 3 does not mediate activation.
SAP binds the cytoplasmic tail of isoform 1 when phosphorylated in the presence of Fyn (in vitro).
SLAMF7 is expressed in spleen, lymph node, peripheral blood leukocytes, bone marrow, small intestine, stomach, appendix, lung, and trachea.
Detected in NK cells, activated B cells, and NK-cell lines; not in promyelocytic, B-, or T-cell lines.
Isoform 3 is expressed at much lower levels than isoform 1.
