Target Information

Lysosomal enzymes containing one or two mannose 6-phosphate (man6P) moieties are transported within the cell via two interconnected cycles mediated by approximately 300 kDa cation-independent mannose 6-phosphate receptors (CI-MPR).
The biosynthetic cycle involves MPR transport of newly synthesized lysosomal enzymes from the trans-Golgi network to late endosomes or early lysosomes. The endocytic cycle carries extracellular lysosomal enzymes from the plasma membrane via clathrin-coated vesicles to early endosomes.
MPRs cycle between these compartments every 3 hours, with most localized in late endosomes, fewer in the trans-Golgi network, and about 10% at the cell surface.
In addition to man6P binding, MPR contains a separate site for type II insulin-like growth factor (IGF-II) and can bind both ligands simultaneously. A soluble ~240 kDa truncated form representing the extracellular domain is found circulating in serum and retains ligand-binding capability.
