Target Information
PUMA는 Bax 및 Noxa와 함께 p53의 전사적 표적이 되는 친사멸성 Bcl-2 패밀리 단백질 중 하나입니다. PUMA 유전자는 BH3 도메인을 포함하는 PUMA-a와 PUMA-b 두 단백질을 암호화합니다. 이 단백질들은 Bcl-2와 결합하고, 미토콘드리아에 위치하며, p53 반응 시 cytochrome c 방출과 세포 사멸을 유도합니다. PUMA는 p53 유도성 세포사멸의 직접적 매개체로 작용할 수 있으며, PUMA 발현 억제 시 p53에 의한 세포사멸 반응이 감소합니다. 따라서 PUMA는 cytochrome c/Apaf-1 경로를 통한 p53 매개 세포사멸에 중요한 역할을 합니다.
