This 22E5 monoclonal antibody reacts with the extracellular domain of mouse Notch1, one of four members of the Notch family of receptors. Notch receptors are 300-kDa single-pass transmembrane proteins. The extracellular domain contains numerous epidermal growth factor-like repeats for ligand binding, while the intracellular domain is involved in cell signaling.
Upon binding its membrane-bound ligand (Delta or Jagged), the Notch receptor undergoes proteolytic cleavage by ADAM-family metalloproteases and gamma-secretase. The released Notch intracellular domain (NICD) translocates into the nucleus, binds RBP-J, recruits co-activators, and activates transcription.
Notch1 is expressed on thymocytes, bone marrow hematopoietic stem cells, T and NK cells, and at lower levels on B cells and monocytes. Certain T-cell acute lymphoblastic leukemia (T-ALL) subsets exhibit Notch1 chromosomal translocation with TCRβ, resulting in constitutively active Notch1. This receptor is involved in T cell lineage commitment, thymocyte development, and Th2 differentiation.

Target Information
The Notch gene family encodes transmembrane proteins with multiple cysteine-rich EGF-like repeats in the extracellular region. Four mammalian Notch genes (Notch1–4) are involved in differentiation of the nervous system and other structures. EGF-like proteins Delta and Serrate act as ligands for Notch1. Mature Notch proteins are heterodimers formed by cleavage of Notch pre-proteins into an extracellular subunit (NEC) and a transmembrane subunit (Ntm).
