Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human Vaspin. Anti-Human Vaspin-specific antibody was purified by affinity chromatography employing an immobilized Human Vaspin matrix.

Sandwich ELISA:
To detect Human Vaspin by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human Vaspin (500-P256Bt) as a detection antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant Human Vaspin.

Western Blot:
To detect Human Vaspin by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. Used with compatible secondary reagents, the detection limit for Recombinant Human Vaspin is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

Vaspin is a newly described adipocytokine expressed predominantly in visceral white adipose tissues. Structural analysis predicts three beta-sheets, nine alpha-helices, and one central loop, characteristic of Serpin family members. Serpins are irreversible serine-protease inhibitors, showing over 30% sequence homology with alpha1-antitrypsin. They play roles in blood coagulation, fibrinolysis, complement activation, angiogenesis, inflammation, and tumor suppression. Vaspin exhibits 40.2% sequence identity with alpha1-antitrypsin, though its protease inhibitory activity remains unknown. Vaspin mRNA expression in visceral fat correlates positively with BMI and body fat percentage. Administration of vaspin to obese mice improved glucose tolerance and insulin sensitivity, normalizing blood glucose levels and modulating diabetes-related adipocytokines such as leptin, adiponectin, resistin, and TNF-alpha. These findings suggest potential clinical applications for vaspin in metabolic syndrome research.
