Target Information
The NF-kB/Rel transcription factors are present in the cytosol in an inactive state, complexed with inhibitory IkB proteins. Activation of NF-kB typically occurs through phosphorylation-induced, proteasome-mediated degradation of IkB. This process involves the high molecular weight IkB kinase (IKK) complex composed of three subunits: IKK alpha, IKK beta (catalytic), and IKK gamma (regulatory). Activation of IKK depends on phosphorylation at Ser177 and Ser181 in IKK beta (Ser176 and Ser180 in IKK alpha), leading to conformational changes and kinase activation.
