Product Specific Information
The MIH4 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the immunoglobulin superfamily. PD-1 includes an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease. PD-1 is expressed predominantly on activated T and B lymphocytes.
Two ligands, PD-L1 (B7-H1) and PD-L2 (B7-DC), have been identified for PD-1. These ligands show overlapping functions and are constitutively expressed on some normal tissues, with upregulation on activated antigen-presenting cells. The MIH4 antibody recognizes a different epitope than clone J105.

Applications Tested:
Tested by flow cytometric analysis of PHA-stimulated peripheral blood mononuclear cells. Recommended usage ≤ 1 µg/test (100 µL final volume). Optimal titration is advised for specific assays.

Target Information
T-cell activation is regulated by inhibitory receptors such as PD-1 (PDCD-1), which mediate signals from antigen-presenting cells (APCs). PD-1 binds two ligands, PD-L1 and PD-L2, and inhibits immune activation in the absence of danger signals (e.g., LPS). PD-1 deficiency leads to hyperactivated immune responses and autoimmune disease. Despite its predicted molecular weight, PD-1 often migrates at a higher molecular weight in SDS-PAGE.
