This 2A5 monoclonal antibody reacts with Foxj1, a 50-kDa member of the Forkhead-box (FOX) family of winged-helix transcription factors. Foxj1 is highly expressed in ciliated epithelial cells and low in naive T and B cells, where activation leads to downregulation. It modulates germinal center formation and Th1 activation by repressing NFκB through IκBβ induction, potentially antagonizing autoimmune reactions like systemic lupus erythematosus and rheumatoid arthritis. Foxj1 is also linked to motile cilia formation in epithelial cells.

Applications Reported: Western blotting, immunohistochemistry (formalin-fixed paraffin embedded tissue), and immunocytochemistry.
Applications Tested: Western blot (mouse tracheal epithelial cells and testes), IHC (mouse lung tissue). Recommended concentration: 1–10 µg/mL, titrate for optimal performance.

Target Information

Forkhead-box J1 (FOXJ1) is a 421-amino acid transcription factor that suppresses T cell activity and spontaneous autoimmunity by repressing NFκB activity. It also inhibits humoral immune responses in B cells. FOXJ1 deficiency results in spontaneous germinal center formation and pathogenic autoantibody development. Abnormal FOXJ1 expression is associated with autoimmune and inflammatory diseases. FOXJ1 is essential for cilia formation and left-right axis determination, increasing calpastatin expression required for basal body anchoring. It may serve as an early marker of epithelial cell differentiation and recovery.
