The 122 monoclonal antibody reacts with mouse B7-DC (PD-L2), a member of the B7 family within the Ig superfamily. B7-DC (~25 kDa) has a short cytoplasmic tail (4aa) and is primarily expressed by dendritic cells and monocytes/macrophages.
Unlike other B7 family members, B7-DC does not bind CD28/CTLA-4 but acts as a ligand for PD-1, regulating T cell proliferation and activation depending on cellular state.
Clone 122 blocks binding of TY25 (Product # 14-5986), another anti-mouse B7-DC mAb.
Recommended for flow cytometric analysis of mouse dendritic cells or B7-DC transfected cells.
Target Information
Programmed death-ligand 2 (PD-L2, B7-DC) is a ligand for PD-1 and part of the inhibitory immune checkpoint pathway.
- Expression: Primarily in dendritic cells; inducible in other immune and non-immune cells.
- Regulation: Upregulated by IL-4, TNF-α, and IFN-γ.
- Affinity: PD-L2 binds PD-1 with 2–6× higher affinity than PD-L1.
- Function: Inhibits T cell activation and cytokine secretion (e.g., IFN-γ, IL-10); can also stimulate proliferation via PD-1–independent pathways.
- Additional receptor: RGMb, involved in respiratory tolerance and BMP/neogenin signaling.
- Human PD-L2 structure: 201-aa extracellular domain, 21-aa transmembrane domain, 32-aa cytoplasmic domain.
