This MA57YW monoclonal antibody recognizes human B7-H7 on live and paraformaldehyde-fixed cells.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated using HDLM-2 cell line; use 5 µL (0.125 µg) per test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test.
Light Sensitivity: Tandem dye sensitive to photo-induced oxidation. Protect vial and samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency.
Excitation: 488–561 nm
Emission: 775 nm
Laser Compatibility: Blue, Green, Yellow-Green lasers
Target Information
B7-H7 (HHLA2) is a membrane protein with three Ig-like domains (IgV-IgC-IgV), unlike other B7 family members that have two. It shares 10–18% amino acid identity and 23–33% similarity with other B7 proteins.
B7-H7 is expressed in epithelial cells of gallbladder, breast, kidney, colon, small intestine, and placenta.
In the immune system, HHLA2 is not constitutively expressed on monocytes/macrophages.
B7-H7 is widely expressed in cancers, notably in triple-negative breast cancer.
