Product Specific Information

This 12.14 monoclonal antibody reacts with mouse Nur77 (NR4A1, TR3, NGFI-B, NAK1), an inducible orphan nuclear receptor expressed in thymocytes and T cell lines. Nur77 promotes apoptosis and plays a role in thymocyte negative selection. It is also critical for steroid biosynthesis in Leydig cells and dopamine-related effects. Interaction with FoxP3 in regulatory T cells has been reported, though not observed with this antibody.

Applications Reported:

  • Intracellular staining followed by flow cytometric analysis.

Applications Tested:

  • Tested in intracellular staining of stimulated mouse thymocytes using Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
  • Recommended ≤0.25 µg per test (100 µL final volume).
  • Cell number range: 10⁵–10⁸ cells/test.
  • Antibody titration recommended for optimal performance.

Light Sensitivity:

  • Protect vial and stained samples from light (photo-induced oxidation).

Fixation:

  • Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark.
  • Clone-specific performance should be empirically determined.

Excitation / Emission / Laser:

Target Information

NR4A1 belongs to the steroid-thyroid hormone-retinoid receptor superfamily. NGFI-B-alpha is expressed in various tissues including brain, adrenal, thyroid, liver, testis, ovary, thymus, muscle, lung, and prostate. It mediates apoptotic signaling and plays a role in hypothalamic-pituitary axis signaling. Translocation from nucleus to mitochondria induces apoptosis. Three transcript variants encode two isoforms; variant (3) or TRC beta differs from variant 1 with a shorter C-terminus. RXR acts as a partner for NR4A1.
