Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. CD14 binds LPS–LBP complexes with high affinity on monocyte surfaces.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells. Recommended at 5 µL (0.25 µg) per test (100 µL final volume).
  • Cell number: 10⁵–10⁸ cells/test (empirically determined)

Fluorochrome Information:
Brilliant Ultra Violet™ 737 (BUV737) emits at 732 nm and is optimized for cytometers equipped with a 355 nm UV laser. Confirm instrument compatibility before use.

Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies (Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use:

  • Super Bright Complete Staining Buffer (SB-4401) or
  • Brilliant Stain Buffer (00-4409)
    to minimize non-specific polymer interactions.

Handling Notes:

  • Light sensitive: protect from light during storage and use.
  • Fixation: compatible with IC Fixation Buffer (00-8222) or 1-step Fix/Lyse Solution (00-5333) for up to 3 days at 4°C in the dark.
  • Methanol tolerance: compatible with short-term methanol fixation (<1 hour). Avoid long-term methanol exposure.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a lipopolysaccharide receptor, binding LPS via LBP. Soluble CD14 (sCD14) discriminates structural differences between LPS molecules and plays a role in neutralizing serum lipopolysaccharides. CD14 also binds apoptotic cells and influences allergic, inflammatory, and infectious processes. Alternative splicing yields multiple transcript variants encoding the same isoform. CD14 dysfunction is associated with mycobacterium chelonae infection and croup.
