Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG (50–80 kDa).
CD16 includes two genes: CD16A (FcγRIIIA) expressed in macrophages and NK cells, and CD16B (FcγRIIIB) expressed in neutrophils.
Two polymorphisms, NA-1 and NA-2, affect phagocytic capacity. CD16 binds IgG immune complexes (preferentially IgG1 and IgG3) and mediates ADCC, phagocytosis, and degranulation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested on human peripheral blood cells (5 µL/test).

Fluorochrome Information:
Brilliant Ultra Violet™ 737 (BUV737) emits at 732 nm, excitable by 355 nm UV laser. Ensure instrument compatibility.

Staining Recommendations:
Use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect vial and stained samples from light.

Fixation:
Samples can be stored up to 3 days at 4°C in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333).
Compatible with short-term methanol fixation (<1 hour).

Target Information

CD16 encodes an Fc receptor for IgG involved in immune complex clearance, ADCC, and viral infection enhancement.
The gene FCGR3A (chromosome 1) is expressed in NK cells, while FCGR3B is expressed in neutrophils.
Mutations are linked to immunodeficiency 20, lupus, and neonatal neutropenia.
Alternative splicing produces multiple isoforms.
