Target Information

RIPK1 is a serine-threonine kinase that mediates inflammatory and programmed necrosis signaling following death receptor ligation, PRR activation, and DNA damage.
Upon TNFR1 activation by TNF-alpha cytokines, TRADD and TRAF2 are recruited to the receptor. RIPK1 phosphorylates DAB2IP at Ser-728 in a TNF-alpha-dependent manner, activating the MAP3K5-JNK apoptotic cascade.
Ubiquitination by TRAF2 via Lys-63-linked chains enhances downstream signaling through the MAPK and NF-κB pathways, leading to inflammatory gene activation. Polyubiquitinated RIPK1 binds IKBKG/NEMO, facilitating NF-κB activation.
RIPK1 interacts with other RHIM-containing adapters to initiate gene activation and cell death, and its association with RIPK3 forms a necrosis-inducing complex.
