Product Specific Information

For reconstitution, add 100 µL distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL (Product #89882).

Target Information

RIPK1 is a serine-threonine kinase that transduces inflammatory and programmed cell death signals following activation of death receptors, pathogen recognition receptors (PRRs), or DNA damage.
Upon TNFR1 activation by TNF-alpha cytokines, TRADD and TRAF2 are recruited to the receptor. RIPK1 phosphorylates DAB2IP at Ser-728, activating the MAP3K5-JNK apoptotic cascade.
Ubiquitination by TRAF2 via Lys-63-linked chains enhances communication with downstream MAPK and NF-κB pathways, leading to activation of inflammatory genes. Polyubiquitinated RIPK1 binds IKBKG/NEMO, a key step for NF-κB activation.
Interaction with RHIM-containing adapters triggers gene activation and cell death. RIPK1-RIPK3 association forms a necrosis-inducing complex.
