Product Specific Information

Store vial at -20°C prior to opening. Aliquot contents and freeze at -20°C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.

Reacts specifically with p53 in human tissues and cell lines. The antibody recognizes a 53 kDa band corresponding to p53. Cross-reactivity with p53 from other sources has not been determined.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. p53 upregulation activates transcription of genes such as p21^waf1^ involved in G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation at Ser15 is critical for p53 activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells, acting as an anticancer agent. It is also linked to aging, oxidative stress, and learning and memory deficits. Normally expressed at low levels, p53 is elevated in transformed cell lines, contributing to malignancy. Mutations in the TP53 gene can lead to loss of tumor suppressor activity and occur in many cancers, including germline mutations in Li-Fraumeni syndrome.
