Target Information
Histone H2A.X (H2AX) is a member of the histone H2A family, one of the four core histones forming the nucleosome. In eukaryotes, DNA double-strand breaks trigger phosphorylation of serine 139 at the carboxy terminus of H2AX, producing γ-H2AX. This phosphorylation can be detected by Western blot or immunofluorescence to assess DNA damage frequency.
Phosphatidylinositol 3-kinases are implicated in H2AX phosphorylation, though it remains unclear if ATM is the primary kinase or if DNA-PK and ATR also contribute. Structurally, H2A.X contains 143 amino acid residues and is synthesized during G1 and S-phase. Its mRNA variants differ in polyA tail presence, with one form behaving as replication-type and the other as basal-type histone mRNA. Histone H2A.X maps to human chromosome 11q23.2–q23.3 and plays a role in chromatin structure and DNA repair through interaction with MDC1.
