This antibody binds to epitopes on the extracellular domains of mouse Fc-gamma-III (CD16) and Fc-gamma-II (CD32), inhibiting their ability to bind antibodies via their Fc region receptors. It also binds the Fc-gamma-I receptor (CD64) via its Fc domain.
This antibody can be used as an Fc-blocking reagent to reduce non-specific staining of cells by antibodies binding through their Fc-region.
The recombinant antibody has been engineered with point mutations that eliminate binding to Fc receptors, enabling reduced background and improved signal-to-noise ratio.

Target Information

CD16 is a 50–65 kDa cell surface molecule existing in two forms: a transmembrane form expressed by natural killer (NK) cells and some T cells, and a phosphatidylinositol-linked form expressed by granulocytes. It acts as a low-affinity receptor for IgG (FcR III) and mediates antibody-dependent cell-mediated cytotoxicity (ADCC).

CD32 is a 40 kDa glycoprotein functioning as a low-affinity receptor for IgG (Fc gamma RII). It mediates endocytosis, secretion activation, cytotoxicity, and immunomodulation. CD32 is expressed by B cells, monocytes, granulocytes, and platelets, and is involved in immune complex phagocytosis and antibody production regulation. Variations in the CD32 gene may increase susceptibility to systemic lupus erythematosus (SLE). Multiple transcript variants encoding different isoforms have been identified.
