Specificity: Human CD34, an approximately 105–120 kDa type I transmembrane glycoprotein expressed on the surface of most human hematopoietic stem and progenitor cells (HSPCs), as well as on mesenchymal stem cells, embryonic fibroblasts, endothelial cells, neurons, and some tumor cell lines.

Target Information

CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) present on various stem cell populations. It serves as a stem cell marker, with reversible expression on human hematopoietic stem cells. CD34 may act as a surface receptor involved in receptor-mediated endocytosis, adhesion, differentiation, and proliferation of hematopoietic stem and progenitor cells.

CD34 expression likely represents a specific developmental state of hematopoietic cells with distinct adhesion and differentiation properties. It may function as an adhesion molecule mediating attachment of stem cells to the bone marrow extracellular matrix or stromal cells, and as a scaffold for lineage-specific glycans that bind to lectins on stromal or marrow components. Additionally, CD34 may present carbohydrate ligands to selectins. The intracellular region of CD34 can be phosphorylated by protein kinase C, suggesting a potential role in signal transduction.

Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
