Product Specific Information

This antibody reacts with the extracellular (N-terminal) domain of standard CD44 (Phagocyte glycoprotein 1), an 80–95 kDa transmembrane glycoprotein (hyaladherin family) present on most cells and tissues including leukocytes, endothelial cells, and mesenchymal cells. It is negative on platelets and hepatocytes.

Western Blot: 60 min incubation on vertical incubator; non-reducing conditions, 6% separating gel.

Target Information

CD44 cell surface antigen is a 100 kDa type I transmembrane glycoprotein widely expressed on human leukocytes, white matter of the brain, and some epithelial cells of the intestine and breast. Several isoforms exist, including the predominant CD44H isoform found in many normal tissues.

CD44 functions as a receptor for hyaluronic acid (HA) and participates in:

• Cell-cell interactions
• Cell adhesion and migration
• Lymphocyte activation, recirculation, and homing

Expression may increase in certain carcinomas and is associated with metastatic potential. CD44 is found in hematopoietic and non-hematopoietic cells, epithelial tissues, and filopodia of cultured keratinocytes. Bone marrow myeloid cells and memory T cells express CD44 at high levels, and peripheral B and T cells can upregulate CD44 expression upon stimulation.

Transcripts undergo complex alternative splicing, generating multiple isoforms with distinct functions. CD44 dysfunction is linked to diseases such as superficial keratitis and lichen sclerosus, and may be related to tumor metastasis formation.
