Product Specific Information

The monoclonal antibody sl6arg recognizes human Arginase-1, a 35 kDa enzyme that converts L-arginine to urea and L-ornithine, aiding nitrogen elimination. Polyamines derived from L-ornithine are crucial for cell proliferation and detoxification from protein degradation.

Expression:
Found in liver, neutrophils, myeloid-derived suppressor cells (MDSC), and neural stem cells. In humans, macrophage expression is undetermined, whereas in mice, M2 macrophage expression is well established. Arginase-1 is commonly expressed in hepatocellular carcinomas and may appear in myeloid populations within breast cancer tumors.

Applications Reported:
Suitable for western blotting, immunohistochemistry (formalin-fixed paraffin-embedded tissues), microscopy, and immunocytochemistry.

Applications Tested:

  • IHC (P): ≤20 µg/mL (tested on human tissues, compatible with low/high pH antigen retrieval)
  • ICC: ≤20 µg/mL (tested on fixed and permeabilized human cells)
    Careful titration is recommended for optimal assay performance.

Target Information

Arginase-1 (Arg1) is a 35 kDa enzyme catalyzing the conversion of L-arginine to urea and L-ornithine, the final step in the urea cycle. Resulting polyamines support cell proliferation and detoxification. By degrading arginine, Arg1 limits substrate availability for NO synthase, reducing nitric oxide production.

Expression Overview:

  • Human and mouse: liver, neutrophils, MDSC, neural stem cells
  • Human: expressed in blood neutrophils, not in CCR3+ granulocytes
  • Mouse: hallmark of M2a macrophages
  • Tumor association: myeloid cells infiltrating tumors, hepatocellular carcinomas
  • Genetic defect: Argininemia (autosomal recessive disorder causing hyperammonemia)
