Product Specific Information
The monoclonal antibody 7F9 (VIIF9) recognizes human, mouse, rat, and avian vinculin and its alternatively spliced isoform, metavinculin. Vinculin is a cytoskeletal protein associated with cell-cell and cell-matrix junctions, involved in anchoring F-actin to the membrane and regulating E-cadherin expression. It binds to talin, paxillin, and alpha-actinin. Dysregulation of vinculin affects cell adhesion, migration, and growth, promoting cancer invasion and metastasis.
Applications Reported:
This antibody has been used in western blotting, immunohistochemistry (frozen and paraffin sections), immunocytochemistry, and microscopy.
Applications Tested:
- Immunocytochemistry on methanol- or formaldehyde-fixed cells ≤ 5 µg/mL
- Immunohistochemistry (formalin-fixed paraffin sections) ≤ 5 µg/mL using low pH antigen retrieval buffer
- Western blot on reduced and non-reduced lysates ≤ 5 µg/mL

Target Information
Vinculin (VCL) is a cytoskeletal protein associated with cell-cell and cell-extracellular matrix adherens-type junctions. It anchors F-actin to the membrane through interactions with talin and alpha-actinin, forming adhesion plaques. Mutations in the VCL gene may cause cardiomyopathy (dilated 1W or familial hypertrophic 15).
