Product Specific Information

The M1/70 monoclonal antibody recognizes mouse CD11b, a 165–170 kDa integrin αM that associates with CD18 to form the αMβ2 integrin (Mac-1). CD11b binds to CD54 (ICAM-1), C3bi, and fibrinogen, and is expressed on macrophages, NK cells, granulocytes, activated lymphocytes, and peritoneal B-1 cells.
M1/70 also cross-reacts with human CD11b, enabling detection on human monocytes, granulocytes, and subsets of NK cells.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes and bone marrow cells at ≤ 0.4 µg/test (100 µL volume, 10⁵–10⁸ cells/test).
Careful titration recommended for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Based on Phiton™ technology utilizing novel nucleic acid dye structures, engineered for optimal fluorescence with reduced spillover and spread.
Excitation: 509 nm; Emission: 665 nm; Laser: 488 nm (Blue).

Target Information

CD11b (Integrin αM, ITGAM) is a 165 kDa adhesion molecule forming a heterodimer with CD18 (β2). Known as Mac-1 or CR3, this complex serves as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X.
Expressed on monocytes/macrophages, granulocytes, activated lymphocytes, subsets of NK and dendritic cells, and microglia.
CD11b expression varies by cell type and tissue, increasing during monocyte maturation.
Dysfunction is associated with systemic lupus erythematosus and ITGAM-related autoimmune susceptibility.
