Product Specific Information

The M1/70 monoclonal antibody reacts with mouse CD11b, a 165–170 kDa integrin alphaM. CD11b associates with CD18 to form Mac-1 (αMβ2 integrin) and binds to CD54 (ICAM-1), C3bi, and fibrinogen.
Mac-1 is expressed by macrophages, NK cells, granulocytes, activated lymphocytes, and mouse B-1 cells.
M1/70 also cross-reacts with human CD11b and can detect this antigen on human monocytes, granulocytes, and NK cell subsets.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes and bone marrow cells, ≤0.4 µg/test (100 µL final volume)

Recommendations:

• Titrate antibody for optimal performance.
• NovaFluor dyes are not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

CellBlox Plus Blocking Buffer Usage:

• Add 5 µL per sample (10³–10⁸ cells).
• Use in all experiments with NovaFluor conjugates.
• For single-color controls, add 5 µL per 100 µL cell sample.

NovaFluor conjugates utilize Phiton™ technology for engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD11b (integrin alpha-M, ITGAM) forms a heterodimer with CD18 (integrin beta-2) known as Mac-1 or CR3.
Expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia.
Functions as receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X.
Expression varies across tissues and increases during monocyte maturation.
Associated with systemic lupus erythematosus and ITGAM-related susceptibility disorders.
