Target Information
The human AF9 gene is one of the most common fusion partner genes with the ALL1 (MLL) gene at 11q23, resulting in the t(9;11)(p22;q23) translocation.
The AF9 gene spans over 100 kb and contains two patient breakpoint cluster regions (BCRs):
- BCR1: within intron 4 (previously called site A)
- BCR2: spans introns 7 and 8
Several structural elements have been identified in AF9, including:
- A colocalizing in vivo DNA topo II cleavage site
- An in vitro DNase I hypersensitive site in intron 7 (BCR2)
- Two scaffold-associated regions (SARs): SAR1 (intron 4) and SAR2 (exons 5–7)
These breakpoint regions share structural similarity with the MLL BCR, suggesting a DNA breakage and repair model for nonhomologous recombination between MLL and AF9.
