Target Information

The human AF9 gene is one of the most common fusion partner genes with the ALL1 gene at 11q23 (also called MLL), resulting in the t(9;11)(p22;q23) translocation.
AF9 spans more than 100 kb and contains two patient breakpoint cluster regions (BCRs):

  • BCR1: within intron 4 (site A)
  • BCR2: spans introns 7 and 8 (site B)

Structural elements identified in AF9 include:

  • DNA topo II cleavage sites and DNase I hypersensitive sites in intron 7 (BCR2)
  • Two scaffold-associated regions (SARs): SAR1 in intron 4 and SAR2 encompassing exons 5–7

These regions share structural similarities with the MLL breakpoint cluster region, supporting a DNA breakage and repair model for nonhomologous recombination between MLL and AF9.
