Target Information
Tau is a neuronal microtubule-associated protein found predominantly on axons. Its primary function is to promote tubulin polymerization and stabilize microtubules. The C-terminus binds axonal microtubules, while the N-terminus binds neural plasma membrane components, suggesting Tau functions as a linker between microtubules and the plasma membrane.
Axonal polarity is influenced by TAU/MAPT localization in the neuronal cell body. Short isoforms contribute to cytoskeletal plasticity, whereas longer isoforms aid in stabilization. In its hyper-phosphorylated form, Tau is the major component of paired helical filaments (PHF), a hallmark of neurofibrillary lesions in Alzheimer’s disease (AD).
Hyper-phosphorylation impairs Tau’s microtubule binding, leading to destabilization and neuronal degeneration in AD. Several kinases phosphorylate Tau, including GSK-3β, PKA, cdk5, and casein kinase II. Hyper-phosphorylated Tau is also found in other neurodegenerative disorders such as Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
