Target Information
Tau는 주로 축삭(axon)에 존재하는 신경세포 미세소관 결합 단백질로, 튜불린 중합을 촉진하고 미세소관을 안정화하는 역할을 합니다.
C-말단은 축삭 미세소관과 결합하고, N-말단은 신경 세포막 구성 요소와 결합하여 Tau가 두 구조를 연결하는 역할을 함을 시사합니다.
짧은 아이소폼은 세포골격의 가소성을, 긴 아이소폼은 안정화에 관여할 가능성이 있습니다.
과인산화된 Tau는 알츠하이머병 뇌의 신경섬유 엉킴(neurofibrillary tangles)을 구성하는 주요 성분으로, 미세소관 결합 기능을 손상시켜 신경세포 퇴행을 유발합니다.
Tau의 인산화는 GSK-3β, PKA, cdk5, casein kinase II 등 다양한 세린/트레오닌 키나아제에 의해 조절됩니다.
과인산화 Tau는 알츠하이머병뿐 아니라 Pick병, 전측두엽 치매, 피질기저변성증, 진행성 핵상마비 등 다양한 중추신경계 질환에서도 발견됩니다.
