Target Information

Tau is a neuronal microtubule-associated protein found predominantly on axons. It promotes tubulin polymerization and stabilizes microtubules. The C-terminus binds axonal microtubules, while the N-terminus binds neural plasma membrane components, functioning as a linker between both. Axonal polarity is influenced by Tau localization within the neuronal cell body.

Tau exists in short and long isoforms, contributing to cytoskeletal plasticity and stabilization, respectively. In its hyper-phosphorylated form, Tau becomes the major component of paired helical filaments (PHF), forming neurofibrillary lesions characteristic of Alzheimer’s disease. Hyper-phosphorylation disrupts microtubule binding, leading to neuronal degeneration. Multiple kinases, including GSK-3β, PKA, CDK5, and casein kinase II, are involved in Tau phosphorylation. Abnormal Tau is also implicated in other neurodegenerative disorders such as Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
