Product Specific Information

The monoclonal antibody eBio84-3C1 reacts with CD43, also known as leukosialin, galactoglycoprotein, and sialophorin. CD43 is a sialomucin with both adhesive and anti-adhesive properties. Expression is found on most leukocytes except resting B lymphocytes. Upon activation, proteolytic processing decreases its surface expression. CD43 is involved in activation of T cells, B cells, NK cells, and monocytes. The counter-receptor for CD43 is CD169/SIGLEC-1, expressed on macrophages.

This antibody is reported for use in flow cytometric analysis.

Applications Tested

Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended use: 5 µL (0.4 µg) per test (100 µL final volume).
Cell number can range from 10⁵ to 10⁸ cells/test.

Important Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• Includes CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample, regardless of the number of NovaFluors used.
  • For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, utilizing novel nucleic acid dye structures engineered for optimized fluorescence with minimal spillover and spread.

Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with a high negative charge, expressed on most hematopoietic cells. It modulates cell adhesion and leukocyte aggregation, interacting with actin-binding proteins (ezrin, moesin) to regulate T-cell morphology and lymphocyte trafficking. CD43 signaling affects LFA-1 adhesiveness and can induce tumor suppressor p53 expression. Dysregulation is associated with Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
