Product Specific Information

The eBioY1/82 antibody reacts with CD68, a member of the sialomucin family related to lysosomal-associated membrane proteins (LAMPs) and scavenger receptors. CD68 is mainly an intracellular protein located in late endosomes but can also be found on the surface of myeloid-derived cells such as monocytes, macrophages, dendritic cells, neutrophils, basophils, mast cells, and some CD34+ progenitor cells. It may play a role in antigen processing, presentation, and lysosomal membrane protection.

Staining with eBioY1/82A shows surface staining on B cells and minor monocyte populations, while intracellular staining targets about 50% of B cells and most monocytes.

Applications Reported: Suitable for intracellular staining followed by flow cytometric analysis.

Applications Tested: Pre-titrated and tested using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol. Recommended use: 5 µL (0.25 µg) per test (100 µL final volume).

Fluorophore Information:
PE-eFluor® 610 is excited by 488–561 nm lasers and emits at 607 nm. Suggested filter: 610/20 band pass (equivalent to PE-Texas Red®). Protect from light due to photo-induced oxidation sensitivity.

Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Target Information

CD68 (Macrosialin) is a 110 kDa integral membrane glycoprotein expressed mainly in lysosomes of monocytes and macrophages, with lower expression in dendritic cells and granulocytes. It is involved in phagocytic activity and lysosomal metabolism. Expression is also observed in certain leukemias and histiocytic conditions, including AML and granulocytic sarcoma.
