Target Information

Junctional complexes between the plasma membrane (PM) and endoplasmic/sarcoplasmic reticulum (ER/SR) are characteristic of excitable cells and mediate communication between cell surface and intracellular ion channels. Junctophilins (JPs) are essential components of these complexes, consisting of a hydrophobic C-terminal segment spanning the ER/SR membrane and a cytoplasmic domain with specific affinity for the PM.

Four JP subtypes have been identified:

  • JPH1: Expressed in skeletal muscle
  • JPH2: Found in all muscle cell types
  • JPH3 and JPH4: Predominantly expressed in brain tissue

In the CNS, JPH3 and JPH4 contribute to subsurface cistern formation in neurons. Mice lacking both subtypes exhibit impaired learning, memory deficits, and abnormal nervous functions.
