Target Information
Junctional complexes between the plasma membrane (PM) and endoplasmic/sarcoplasmic reticulum (ER/SR) are essential in excitable cells, mediating communication between surface and intracellular ion channels.
Junctophilins (JPs) are key structural components of these complexes, containing a hydrophobic C-terminal membrane-spanning region and a cytoplasmic domain with PM affinity.
Four JP subtypes are known:
- JPH1: Expressed in skeletal muscle
- JPH2: Found in all muscle cell types
- JPH3, JPH4: Predominantly expressed in the brain
In the CNS, JPH3 and JPH4 contribute to neuronal subsurface cistern formation. Mice lacking both subtypes exhibit impaired learning, memory, and abnormal nervous function.
