Target Information

Junctional complexes between the plasma membrane (PM) and endoplasmic/sarcoplasmic reticulum (ER/SR) are characteristic of excitable cells, mediating cross talk between cell surface and intracellular ion channels.
Junctophilins (JPs) are key components of these complexes, composed of a carboxy-terminal hydrophobic segment spanning the ER/SR membrane and a cytoplasmic domain with affinity for the PM.

Four JP subtypes have been identified:

  • JPH1: Expressed in skeletal muscle
  • JPH2: Found in all muscle cell types
  • JPH3, JPH4: Predominantly expressed in the brain

In the CNS, JPH3 and JPH4 contribute to subsurface cistern formation in neurons. Mice lacking both subtypes show impaired learning, memory, and abnormal nervous functions.
A repeat expansion in JPH3 is associated with Huntington disease-like 2. At least two isoforms of JPH3 are known.
