Product Specific Information

This antibody recognizes CBX5 (Chromobox protein homolog 5) and binds to a folded domain (amino acids 18–75).
CBX5 is a component of heterochromatin that recognizes and binds histone H3 tails methylated at Lys-9 (H3K9me), leading to epigenetic repression.


Target Information

Heterochromatin protein-1 (HP1) is a methyl-lysine binding protein localized at heterochromatin sites, mediating gene silencing.
Mammalian methyltransferases that methylate histone H3 on lysine-9 generate a binding site for HP1 proteins, which act as adaptor molecules in chromatin structure and gene repression.
High-affinity recognition of methylated histone H3 by HP1 requires a functional chromodomain, serving as a specific interaction motif for the methyl epitope on lysine-9.
In vivo, heterochromatin association of HP1 proteins is lost in Suv39h double-primary mouse fibroblasts but restored after reintroduction of catalytically active SUV39H1 HMTase.
