Product Specific Information
The 4S.B3 monoclonal antibody reacts with human interferon-gamma (IFN-γ), a 17 kDa cytokine produced by activated T and NK cells with antiviral and antiparasitic properties. IFN-γ synergizes with TNF-α to inhibit proliferation of normal and transformed cells and exerts immunomodulatory effects on various cell types expressing high-affinity IFN-γ receptors. Glycosylation does not affect its biological activity.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and validated for intracellular staining of stimulated human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and standard protocols. Recommended use: 5 µL (1.0 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Brilliant Ultra Violet™ 395 (BUV395) emits at 395 nm and is designed for cytometers equipped with a 355 nm ultraviolet laser. Verify instrument compatibility before use.
Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies (e.g., Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Target Information
Interferon gamma (IFN-γ, Type II interferon) is a macrophage-activating and immune interferon produced primarily by T-lymphocytes and NK cells in response to antigens and cytokines. It is a dimeric glycoprotein (~45 kDa) comprising two 146 amino acid subunits. IFN-γ exhibits antiviral, antiproliferative, and immunomodulatory activities, including induction of MHC I/II, macrophage activation, and enhanced immunoglobulin secretion by B cells.
Human IFN-γ is species-specific and does not cross-react with mouse IFN-γ. Activation occurs via IFN-γ receptor I/II and the JAK-STAT pathway. Gene mutations are associated with conditions such as aplastic anemia.
