Product Specific Information

Description:
The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells, and binds glycosylated, fucosylated, and sulfated sialylated glycoproteins such as CD34, glycam-1, and MAdCAM-1. These interactions mediate lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) in peripheral lymphoid tissues.

Applications Reported:
Flow cytometric analysis

Applications Tested:
Pre-titrated and tested for flow cytometric analysis of normal human peripheral blood cells.
Recommended use: 5 µL (0.2 µg) per test (100 µL final volume).
Cell number: 10⁵–10⁸ cells/test (empirically determined).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability detection.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor of the selectin family.
It consists of:

• Aminoterminal C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Spacer, transmembrane, and cytoplasmic regions

CD62L is constitutively expressed on most leukocytes (lymphocytes, monocytes, polymorphonuclear cells) except for a subset of memory T cells.
