Product Specific Information

The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, and sulfated sialylated glycoproteins such as CD34, GlyCAM-1, and MAdCAM-1, mediating lymphocyte rolling and homing to high endothelial venules (HEV).

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells; use at 5 µL (1 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

NovaFluor Compatibility:
Not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.

CellBlox Plus Blocking Buffer:
Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates use Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures minimizing spillover and spread.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor with:

• Aminoterminal C-type lectin domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Transmembrane and cytoplasmic regions

It is constitutively expressed on all classes of leukocytes including lymphocytes (except many memory T cells), monocytes, and polymorphonuclear cells.
