Product Specific Information

The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, sulfated sialylated glycoproteins such as CD34, glycam-1, and MAdCAM-1, mediating lymphocyte rolling on activated endothelium and homing to peripheral lymphoid tissues.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (0.2 µg) per 100 µL cell sample (10⁵–10⁸ cells/test).

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample before combining antibodies with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor of the selectin family. It includes:

• C-type lectin domain
• EGF-like domain
• Two SCR sequences
• Transmembrane and cytoplasmic regions

CD62L is constitutively expressed on most leukocytes including lymphocytes, monocytes, and polymorphonuclear cells, except a subset of memory T cells.
