Product Specific Information

The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, and sialylated glycoproteins such as CD34, glycam-1, and MAdCAM-1, mediating lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometry on normal human peripheral blood cells at 5 µL (0.2 µg) per test.
A test is defined as the amount of antibody that stains a cell sample in 100 µL final volume (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor. It includes:

• C-type lectin binding domain
• EGF-like domain
• Two SCR sequences
• Spacer, transmembrane, and cytoplasmic regions

Expressed on all leukocytes including lymphocytes (except many memory T cells), monocytes, and polymorphonuclear cells.
