Product Specific Information

The monoclonal antibody eBio8D9 reacts with an epitope shared by mouse CD21 (CR2) and CD35 (CR1). These proteins are alternatively spliced transcripts of the Cr2 gene, producing surface proteins of 145 and 190 kDa.
CD21 and CD35 are expressed by mature B cells, but not on thymocytes, peripheral T cells, erythrocytes, or platelets. Some evidence suggests expression on macrophages. CD21 acts as a receptor for complement component C3d and Epstein-Barr virus (EBV). Together with CD19 and CD81, CD21 participates in B-cell activation through the B cell receptor.
Cr2-deficient mice display impaired inflammatory and humoral immune responses in vivo.
The anti-mouse CD21/CD35 monoclonal antibody clones eBio4E3 and eBio8D9 do not cross-block each other, indicating distinct epitope binding.

Applications Reported: Flow cytometric analysis and immunoblotting (WB). Also suitable for functional assays to crosslink CD21/CD35 to itself or sIgM.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes at ≤0.5 µg/test. A test is defined as the amount of antibody staining a cell sample in a final volume of 100 µL. Cell numbers can range from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information

CD21 (Complement Receptor 2, CR2):
Binds C3 complement fragments and plays roles in immunocomplex uptake, memory B cell survival, and humoral response development. Serves as a receptor for Epstein-Barr virus and is involved in prion targeting to follicular dendritic cells. A soluble form (sCD21) retains ligand-binding ability.

CD35:
Encodes a membrane glycoprotein found on peripheral blood cells, glomerular podocytes, and follicular dendritic cells. Functions as a receptor for complement components C3b and C4b, regulating complement cascade activity. Variations form the basis of the Knops blood group system. A secreted plasma form has been described, though its full-length nature is undetermined.
